Markers and Staining Protocols Pig Monocytes

Using monoclonal antibodies and flow cytometry, monocytes in the pig are mostly identified with antibodies to CD172a, but granulocytes, which are also positive, have to be excluded by initial PBMC isolation and/ or via light scatter. Also, CD14 expression has been used for this purpose, which at the same time can serve to define the high and low subsets. These markers are typically combined with CD163 to define the CD163 high and the CD163low subsets. A typical example of such a pattern in shown in the figure below.

Percoll-isolated blood monocytes from Large-White pigs were stained with anti-CD172a (BA1C11, IgG1),
anti-CD14 (MIL-2, IgG2b) and anti-CD163 (2A10, IgG1) mAbs.
